Limited Face-to-Face Human Subject Interaction This research includes face-to-face human subject interaction. This request is for IRB protocol #1617405-2. We already have approval for consenting and screening for this study. The purpose of this study is to assess the impact of dietary sodium and fructose on blood pressure and inflammation in healthy adults 18-45 years old. We now request permission to begin data collection.

This data collection involves 6 visits to the lab. The participant will come to the lab before and after 3 different 7 day diets, separated by ~30 days.
The first visit in each condition will consist of subjects briefly obtaining materials required for the week. The second follow-up visit (2 hours in length) will involve an in-lab data collection and follow the outline detailed below. Dietary counseling for intervention diets will be provided to the participant virtually unless the participant requests a face-to-face visit. If face to face visits are requested, they will occur in a conference room where greater than 6 feet of physical distance can be maintained. All materials (BP cuff, 24-hour urine container, actigraph) will be thoroughly cleaned, sterilized, and prepared in separate packaging for the study participants. All data collection visits following the 7-day conditions will be performed identically and include the procedures described below.

In lab data collection study visit (2 hours in length) separated by ≥ 1 month (3 per subject)

• Weight, urine collection, and materials return: 1 min of close contact while exchanging materials and collecting measurements (5 mins). (1 researcher needed)

• Subject rest period in supine position: 30 min duration (1 researcher needed + 1 researcher in non-close contact)

• Heart rate and blood pressure (placement and removal of 4 sticker electrodes placed on subject’s chest to monitor heart rate, and the blood pressure cuff on subject’s arm): 3 min of close contact during 15 min of measurement duration (1 researcher needed + 1 researcher in non-close contact).

• Catheter placement for blood draw and obtaining venous cells: 15 min of close contact involving a research RN plus 2 researchers. One of the researchers is not in close contact.

• Ultrasound measurement of brachial artery (set up & clean up): 15 min of close contact during 30 min of measurement duration (1 researcher needed + 1 researcher in non-close contact)

• Central BP Measurements: placement and removal of UsCom BP cuff on subjects arm to measure central BP 2 min of close contact during 5 mins of measurements (1 researcher needed + 1 researcher in non-close contact).

Blood and urine samples are processed in the wet lab (STAR 128). Multiple labs utilize this space. A Google calendar is used to schedule the space, keeping in mind the required personnel density per square foot. Physical distance will be maintained.

Proper PPE will be worn for all participant encounters. Frequent hand washing is performed. We have a large team, but team members work on different tasks and not all together (e.g., consenting subjects, diet counseling, data collection, data analysis). Team members also do not all come to building on the same days.

All lab personnel complete the UD daily symptom check. All lab personnel have already completed the UD COVID training, and have reviewed COVID-related safety procedures.

All research participants will complete the standard STAR building screening check upon entering the building (temperature check, etc.).

Physical Distancing and Density Requirements For our wet lab, a Google calendar is used to schedule the space, keeping in mind the required personnel density per square foot. Physical distance will be maintained. Team members also do not all come to building on the same days. When research participants are being tested, only the required personnel are in the room. Distance is maintained at all times except for when procedures are being performed (details above for each procedure).
